When measuring the spacing of two identical holes to be able to add a third at the same spacing, just measure from one edge of the first hole to the same edge of the second hole. it's the same measurement as center to center, but much easier to get accurately.

...just don't forget that the mark you made wasn't the C/L of the hole when you drill!

For that measurement, add 1/2 the diameter of the hole to the above measurement. Again, this only applies to holes of the same diameter.
